Development of an Enhanced High-Yield Influenza Vaccine Backbone in Embryonated Chicken Eggs

Vaccination is an efficient approach to preventing influenza virus infections. Recently, we developed A and B vaccine backbones that increased the yield of several viruses in Madin–Darby canine kidney (MDCK) African green monkey (Vero) cells. These also viral replication embryonated chicken eggs, which are most frequently used platform for manufacturing. In this study, further increase titers introduced random mutations into ‘internal genes’ (i.e., all genes except those encoding hemagglutinin neuraminidase proteins) high-yield backbone previously. The randomly mutated were sequentially passaged eggs select variants with replicative ability. We identified a candidate conferred higher growth than parental backbone. Although observed increases may be considered small, they highly relevant manufacturers.
